Effect of Estrogen on Antioxidant Enzymes on Skeletal Muscle in Oophorectomized Rats

Abstract
The purpose of this study is to evaluate and antioxidative effect of estrogen on the skeletal muscle in oophoretomized rats. Thirty Sprague-Dawley rats of three months age were equally divided into 3 groups ; group 1 as control group with sham operation, group 2 as experimental group with oophorectomy and group 3 as oophorectomized group treated with estrogen. Estradiol 5 §·/§¸ BW was administered three times per week for sixth weeks after oophorectomy. Soleus and gastrocnemius were obtained to measure the amount of protein carbonyls and thiobarbituric acid reactive substances(TBARS) as indices of oxidative stress and the activities of antioxidant enzymes. The differences in the amounts of protein carbonyls in muscle tissues were not significant among all groups. However, the level of TBARS in soleus was higher in group 2 than that in group 1. Its increase was enforced by estrogen treatment. in gastrocnemius, the levels of TBARS showed a similar pattern to those in soleus, though without statistical significance. These results indicate that esgrogen seems to have oxidative characteristics rather than antioxidative effect on both types of the skeletal muscle. The activities of antioxidant enzymes were higher in soleus than in gastrocnemius. However, estrogen treatment resulted in different effects on antioxidant enzymes, showing an increase in activities of glutathione reductase and perxodase of soleus and a decrease in activity of catalase of gastrocnemius. In summary, estrogen increased the level of oxidative stress in skeletal muscle probably due to an oxidative property of estrogen and also modulation of antioxidative enzymes. The differences of estrogen responses are considered to be dependent on muscle fiber type.
